Plasmid Mapping:

This is a useful technique where if one knows the total number of base pairs in a plasmid, upon digestion with single and multiple restriction enzymes their order of occurance may be elucidated by analyzing the sizes of the resulting bands. This step is often a precursor to base sequencing.
